How and where can i define project in LN 6.1 puchase order line.

Still on the PO line. In case you use session tdpur4100m900 you can add the project related fields to the grid (personalize grid option) or you can use the detailed lines session tdpur4101m000.

For Standard Item you can not define project in Purchase Order, however for Customize item Project field will automatically appear.

This is not true also standard items can be purchased with the project data being populated.
The only prerequisite is that Item project data exist when populating the project field.
In case of project specific items (the project part of the item code populated) indeed the standalone project field is automatically populated.
